Predict your bioprocess
BioProGuide is a free, browser-based tool for mechanistic process prediction in biologics development. It is designed for students, academic laboratories, and small biotechnology companies that do not have an in-house process development team. The tool provides end-to-end guidance for recombinant protein production, starting from an amino acid sequence and concluding with formulation recommendations, without requiring deep prior expertise in bioprocessing. The workflow begins when a user inputs an amino acid sequence and specifies the intended use case and route of administration. BioProGuide analyzes the protein's intrinsic properties, including isoelectric point, molecular weight, hydrophobicity, disulfide bonds, post-translational modification sites, and stability hotspots. This analysis informs subsequent decisions. The tool scores potential expression hosts based on PTM capability, folding, proteases, codon usage, and capacity, generating a producibility score with rationale. It then guides the design of the expression construct, covering codon-optimized coding sequences, promoters, signal peptides, tags, and copy number. The tool models upstream process kinetics and downstream purification steps, such as affinity capture, polishing chromatography, refolding, tangential flow filtration, and viral filtration, using mechanistic column sizing. Formulation guidance is provided based on predicted folding and surface motifs. Typical use cases include generating a first mechanistic plan for a new target, making early go or no-go decisions before budget commitment, and establishing a formulation starting point before engaging a contract development and manufacturing organization. It is also used to create defensible rationale for investor updates, provide first-pass feasibility data for grant proposals, and screen host and strategy options before pilot studies. In academic settings, the tool serves as a training resource for new students, a source of cited rationale for reviewer panels, and a platform for bridging textbook theory with real-world production decisions. Users can run live demonstrations, adjusting parameters to observe how changes affect the entire pipeline. BioProGuide connects decisions across traditionally siloed planning stages, ensuring that each step constrains and informs the next. It provides a comprehensive view of the pipeline, from construct design through formulation, allowing users to defend their choices with a clear rationale. The tool is accessible entirely through a web browser and requires no software installation.
